檢驗原理 單純皰疹病毒2型IgM免疫診斷試劑盒

用抗原包被微量板孔，制成固相載體。加患者血清到板孔中，其所含的抗體特異性地與固相載體中現存抗原結合，形成免疫復合物。除去多余物質后，加入結合了堿性磷酸酶的IgG、IgA或IgM抗體，使之與上述免疫復合物反應。洗板，除去多余的結合物，加入底物（對硝基苯磷酸鹽）。其與酶結合的免疫復合物反應，產生有顏色產物，顏色強度與特異性抗體含量成正比。

HSV具有典型皰疹病毒形態特征。根據生物化學、生物學、流行病學等分為兩個血清型，即HSV-1和HSV-2。二者基因組相似，序列有50%的同源性，通過DNA限制性內切酶分析來區分。HSV基因組大約152kb，34個基因，編碼70多個多肽（polypeptides）。特別是?基因編碼的晚期蛋白中有11種包膜糖蛋白（gB、gC、gD、gE、gG、gH、gI、gJ、gK、gL、gM），有些功能較清楚。其中gB和gD與病毒吸附和穿入有關，是與細胞特異性受體相互作用的病毒配體分子。gD誘導產生中和抗體的能力zui強，可用于研制疫苗。gC是補體C3b―結合蛋白（complement C3b-binding protein）。gE是Fc受體，可與IgG的Fc端結合。gG為型特異性抗原，以此抗原能區別HSV-1（gG-1）和HSV-2（gG-2）。gH與病毒的釋放有關。
HSV對動物感染宿主范圍較廣。常用實驗動物為家兔、豚鼠及小鼠等。HSV在多種細胞中能增殖，常用原代兔腎、人胚腎細胞以及地鼠腎等傳代細胞培養分離病毒。感染細胞很快出現明顯細胞病變，并出現嗜酸性核內包涵體。
