一、非特異性染色的主要因素
組織的非特異性染色的機(jī)理很復(fù)雜，其產(chǎn)生的原因主要可分為以下幾點：
（1）一部分熒光素未與蛋白質(zhì)結(jié)合，形成了聚合物和衍化物，而不能被透析除去。
（2）抗體以外的血清蛋白與熒光素結(jié)合形成熒光素脲蛋白，可與組織成分結(jié)合。
（3）除去檢查的抗原以外，組織中還可能存在類屬抗原（如Forssman氏抗原），可與組織中特異性抗原以外之之相應(yīng)抗體結(jié)合。
（4）從組織中難于提純抗原性物質(zhì)，所以制備的免疫血清中往往混雜一些抗其他組織成分的抗體，以致容易混淆。
（5）抗體分子上標(biāo)記的熒光素分子太多，這種過量標(biāo)記的抗體分子帶過多的陰離子，可吸附于正常組織上而呈現(xiàn)非特異性染色。
（6）熒光素不純，標(biāo)本固定不當(dāng)?shù)取?nbsp;
First, non-specific staining of the main factors
Tissue non-specific staining mechanism is very complex, its causes can be divided into the following points:
(1) A portion of the fluorescein is not bound to the protein, forming a polymer and derivative that can not be removed by dialysis.
(2) Serum proteins other than antibodies bind to fluorescein to form fluorescein urea, which binds to tissue components.
(3) In addition to the antigen to be examined, there may also be a generic antigen (such as Forssman's antigen) in the tissue that binds to a corresponding antibody other than the specific antigen in the tissue.
(4) It is difficult to purify the antigenic substance from the tissue, so the prepared immune serum is often mixed with antibodies against other tissue components, so that it is easy to be confused.
(5) There are too many fluorescein molecules labeled on the antibody molecule. This excess labeled antibody molecule with too many anions can be adsorbed on normal tissues and present nonspecific staining.
(6) impure fluorescein, specimen fixed improperly.
